      Immunohistochemicaltechniqueshavebeeninuseasadjunctivemethodsfortherecognitionofcellsintissuesectionsforatleast50years.Thefirstmethodreporteduseoffluorescentlabelsin19421.In1974,reportsofusingenzymaticlabels,suchashorserADIshperoxidase(HRP)inroutineH&Eparaffinsectionsappeared2,3.Themethodshavesincebecomethe"standardofcare"insurgicalpathologywhenclassicmethodsalonefailtoyieldadefinitivediagnosis4-9.

      Earlytechniqueswerebasedontheperoxidase-anti-peroxidase(PAP)reaction28,whileimprovementsexploitedthestrongaffinityofavidinforbiotin10.ThistechniqueusesHRP-labeledstreptavidin.Thesecondaryantibodyisconjugatedwithbiotinandthestreptavidin-HRPcomplexreactswiththebiotinonthesecondaryantibodies.Theresultingbiotin-avidin-HRPcomplexcanreactwiththeprimaryantibodyboundtothespecificepitopeofthetargetantigen.TheHRPenzymesofthestreptavidincomplexthencatalyzethesubstrate/chromogenreactiontoformacoloredreactionproduct(browntoblackwhenusingDABasthechromogen)attheantigensite.ABIOLOGicalstainisthenusedtovisualizethewholetissuesection.

      ·Diaminobenzidine(DAB)maybecarcinogenic.Thissolutionmaycauseirritationuponskincontact.Avoidinhalation,ingestion,orskincontactandweargloveswhenhandlingDAB.Ifskincontactoccurs,flushaffectedareawithwater.Spillsmaybe"neutralized"withbleach25,26.Disposeofaccordingtolocalregulations.

      ·Inhalationoringestionofxyleneorthehighlyallergenicfixativeformaldehydeisharmful.Thesematerialsshouldbehandledwithgloves.Prepareinafumehood.Ifskinoreyecontactoccurs,washthoroughlywithcopiousamountsofwater.
